something in the combination of your site’s JS must throw Safari off, try excluding some of the JS from JS optimization, starting broad (e.g. wp-content/plugins or wp-content/themes or wp-includes) and then narrow things down?

Well, disable AsyncJS for now and focus on AO first maybe. Start by adding one of those strings (wp-content/plugins or wp-content/themes or wp-includes) to the comma-separated JS optimization exclusion field on Autoptimize’s main settings page and see with which one things work/ break.
