Unable to use support theme

i can’t understand how to add theme support. Can you plz explain step by step to add theme support. i have tried this but not able to understand that how to create single-ex_test.php with my theme html structure.
can you explain in detail that how do we do this step. if possible plz share the video for tutorial purpose.
