Yes, everytime while upgrading the plugin, entire old directory will be deleted and will be replaced by new updated directoy. This means, if you have included translation file inside plugin directory, It will be deleted too. The correct place to store translations is wp-content/languages/plugins/.

That’s what I did, hence my question about how (or when) the translation can be ‘officially’ included in a release. Should the translations be first reviewed by someone ? Who ? And how can I contact that reviewer ?

Once translated, the translated string will be in pending status. They need to be approved by Translation admin for that language.
