where do I need to start to troubleshoot this?

Try re-saving your permalink structure at Settings/Permalinks in your admin panel. If WordPress cannot automatically edit the .htaccess file, it will provide manual instructions after saving.

Just tried that, nothing changed, unfortunately.

Are you using some sort of special search function, plugin, or special theme-based search function? The search query URLs should be ending with /?s=term and not /search/term/ like yours.

I have the Enhanced Search Box plugin installed. When I deactivate it, however, the search still does not work…it never did, actually.

Try decativating all plugins. If that resolves the issue, reactivate each one individually until you find the cause.

If that does not resolve the issue, try switching to the Twenty Ten theme to rule-out a theme-specific issue.

If that does not resolve the issue, try switching to the Twenty Ten theme with all plugins deactivated.
