MLA does not have any features that alter the files attached to Media Library items in any way. This was an early and explicit decision on my part because I didn’t want to run the risk of damaging anyone’s files.
However, you can use MLA to add GPS information to custom fields or Image Metadata elements (in the WordPress Image Metadata database field, not the image file) assigned to your items and then use the information in your application. Here are two earlier topics that might give you some ideas:

Those topics make use of EXIF information embedded in the image files. For your application the solution is a bit more involved. Rather than using a template such as template:[+exif:GPS.Latitude+]
, you would leave the Data Source set to “– None (select a value) –“. If you check the Quick Edit and Bulk Edit boxes you can enter the GPS values manually in the Media/Assistant Quick or Bulk Edit areas. You would have to make sure the format is correct, but at least the Bulk Edit area would let you update several images at once.
You can see in the earlier topics that many mapping plugins use the Image Metadata area to store their data and there are different formats from one plugin to the next. Your choice of storing the information in the Image Metadata or in separate custom fields will be influenced by your specific application.
