Misled links on blog

I have created a website on WordPress and the front page is a static page of my home page. I set the posts page to blog page. I just created my first post today and the permalink is mydomain.com/post. When I click it, the page the post is suppose to be on is a 404 error… I tried altering the permalink so that it said mydomain.com/blog/post but I couldn’t. I’ve tried theme options and my file manager to alter this issue but I just can’t. Is there anyone who can fix my issue? The post shows on the blog page but when I click the excerpt it brings me to that error, thanks.
