Latest update blocks backend access for some users and do you respond to email?

After several runs of testing, the newest update can be confirmed to be the culprit in completely blocking access to the backend for certain user roles who have previously been set up to have full access.
What can be done to fix this?
Also, 17 days ago I sent an email to your listed IT on your website after you referred me there in the forums here. In the forums I also replied with a question to something rather vague that you mentioned, but no one has responded in either of these places. 17 days….
