.htacces for Permalink structure problems

I’m trying to abilitate Permalink structure and have created the .htaccess file with the chmod 666 but when trying it my server gives me a 500 error.
Is this a problem with my server configuration that doesn’t let me use permalink structure?
Would I need to check this with my ISP?
