Do we need to keep this plugin enabled, as opposed to your Index WP MySQL plugin which can just be run once and turned off?

Yes, this plugin needs to stay activated to work properly.

    Why?

It hooks the functions that add / change / delete users from your system so it can maintain its fast-access user metadata.

And, upon deactivation it removes that metadata … on a site with thousands of users, it’s just rude to leave an extra row in wp_usermeta on deactivation, so I didn’t do that.

(Some plugins remove their data upon deletion. But this one removes it on deactivation. If it’s not active the metadata will get stale fast.)

Thanks! I figured as much. And that’s very courteous of you to clean up after yourself ??

My site is still under development, so I can’t actually test anything. Though I expect a lot of users when it is live. So, one other question:

Does it only help with user stuff in the admin? Or, if you were querying many/all users elsewhere on the site – be it a front end member directory or whatever – would this help as well?
